This site has been developed to facilitate distribution of tomato cDNA and BAC
clones developed with support from the
NSF Plant Genome Program and as part of the International SOL project focused
on tomato genome sequencing. As we are not supported for resource distribution and requests
are extensive this is being done on a cost recovery basis. Additional information regarding
tomato ESTs, fosmid and BAC clones and libraries can be found at the
Solanaceae Genomics Network (SGN).
IMPORTANT
A number of important points should be kept in mind when ordering tomato cDNA or BAC clones.
As is typical for all large scale clone programs there are errors in annotation. Due to a
combination of human and machine errors, the clone IDs found in SGN, TIGR, GenBank or other
databases may not always match the indicated sequences. As a result, if you order clone X and
sequence it you may find it is not what you wanted. For example, in the tomato EST collection
we estimate an average error rate of approximately 10% (though this can be significantly higher
in specific libraries or segments of libraries). You should account for this error in your
clone requests and this site helps you to do so in several ways. First, a subset of clones
including those used in the microarray have been re-sequenced and are thus the most accurate
(though still not 100%). Our clone search tool will assist you in identifying clones from this
subset that match your sequence of interest (You can blast against the whole microarray
re-sequenced collection from here or against all
available full BAC, BAC end
or fosmid end sequences at SGN). We also strongly suggest that you request additional
clones from the EST contig if available (you can go to SGN to get additional member information) and especially if re-sequenced
clones are not available.
You should verify any clones you receive via sequencing (to verify BAC or fosmid end sequence
or to verify cDNA insert sequence). Clones that are not as expected cannot be replaced
for free unless error on our part is apparent.
